ABSTRACT

Objective: To investigate the effect on the proliferation, mineralization and adipogenic differentiation of dental pulp stem cells ( DPSCs) under different oxidative stress by p38-MAPK and STAT3 signaling pathway. Methods: DPSCs was isolated and identified from pulp tissue by trypsin digestion. The CCK8 detection cell model was divided into oxygen group, 3% O2 group, 6% O2 group, 9% O2 group firstly, each group included the SH-4-54 treatment group, and the SB203580 treatment group, a total of 12 groups. Cell proliferation was detected by CCK8 test for 6 h, 12 h, 24 h, 36 h, 48 h. We observed the differentiation of DPSCs mineralization by alizarin red staining involved in p38-MAPK and STAT3 signaling pathway, and the a-dipogenic differentiation of DPSCs by oil red O staining. Results: DPSCs were cultured by the enzyme digestion method, flow cytometry showed that DPSCs spec-ific marker Stro-1 expression was 96. 85%, CD146 positive expression was 90. 94%, the positive low expression of CD45 was 1. 02%, the positive low expression of CD34 was 40. 76%, p38-MAPK inhibited DPSCs with different oxygen environment. The proliferation of STAT3 promotes the survival of groups DPSCs, the p38-MAPK signaling pathway plays a positive role in the differentiation of mineralization of DPSCs and the STAT3 signaling pathway plays negative regulation role, the two signaling pathway play opposite effects in adipogenic differentiation research. Conclusion: The proliferation rate of DPSCs is higher in normoxic environment, and the STAT3 signal pathway can promote the proliferation of DPSCs, the MAPK signal pathway promotes the odontoblastic differentiation of DPSCs.

ABSTRACT

@#AIM: To evaluate the changes in intraocular pressure(IOP)after laser suture lysis after trabeculectomy associated with 5-Fluoroaracil and the correlate the effect in relation to the time of laser suture lysis.<p>METHODS: Prospective interventional study of sixty eyes of sixty patients. Trabeculectomy associated with 5-Fluoroaracil was performed for 60 patients with glaucoma. Laser suture lysis was carried out according to the IOP of post trabeculectomy. 21mmHg was used as the cut off for the achievement of target IOP. According to the timing of laser suture lysis, patients post trabeculectomy were divided into three groups: <2wk, 2<sup>nd</sup>-4<sup>th</sup>wk and >4wk.<p>RESULTS: The differences of IOP prelaser suture lysis showed no statistical significance among three groups(<i>P</i>=0.999). The difference of IOP following 12wk post laser suture lysis showed significant statistical significance(<i>P</i><0.05)among three groups. When laser suture lysis was performed during 2<sup>nd</sup>-4<sup>th</sup> week post trabeculectomy, 28 patients(74%)achieved the target IOP. Target IOP achieved is negatively correlated to higher week of lysis(<i>P</i><sub>trend</sub>=0.069).<p>CONCLUSION: Laser suture lysis during 2<sup>nd</sup>-4<sup>th</sup> week post trabeculectomy associated with 5-Fluoroaracil is an effective and safe technique for reducing IOP.

ABSTRACT

? AlM: To observe the clinical curative effect of intravitreal Bevacizumab injection combined duplex technique in treatment of neovascular glaucoma ( NVG) . ?METHODS:Totally 25 eyes of 25 patients with NVG who underwent intravitreal Bevacizumab injection of 1. 0mg (0. 05mL), after the regression of iris neovascularization, 5 eyes with anterior chamber paracentesis fluid auxiliary controlled intraocular pressure. After 2wk, patients were treated by trabeculectomy and phacomulsification (9 eyes were implanted intraocular lens ) . The changes and complications of intraocular pressure, visual acuity, corneas and neovessels were observed after surgery, and followed up 12mo. ?RESULTS:After injection Bevacizumab in 25 eyes, iris neovascularization of 20 eyes subsided in 3 ~ 5d, and 5 eyes subsided in 7d. After controlling intraocular pressure, count of the corneal endothelial cell were 1 629±226mm2 , and none suffered decompensation of corneal endothelium after two- surgery of trabeculectomy and phacomulsification. After followed up 12mo, intraocular pressure of 20 eyes were controlled in normal range; 2 eyes could control in normal range after treated by a kind of anti-glaucoma medicine and 3 eyes was 34 ~38mmHg after treated by anti- glaucoma medicine. 9 eyes had improved vision after implanted intraocular lens. ?CONCLUSlON:lntravitreal Bevacizumab injection can subside iris and anterior chamber angle neovascularization effectively in a short time and reduce intraocular pressure. lt can also reduce the risk of bleeding during operation or after operation. lntravitreal Bevacizumab injection combined with two- surgery of trabeculectomy and phacomulsification can treat neovascular glaucoma effectively.
